Each compressed file (.zip file) name corresponds to the measurement sample (the variant name, and if present, the chaperone name). After uncompressing the archives, various folders are generated: - the "Raw data files" folder contains the acquired photon stream files in .ht3 format. - For all the FRET measurements presented in the manuscript, a folder named "BIN files - IPT-0p05-0p05-N-40" is also provided. It contains the analysed burst files with a maximum interphoton time of 0.05 ms, minimum interphoton noise of 0.05 ms, and a minimum total number of 40 photons. All the other settings used for analysis are mentioned in the "Settings.txt"*. - In the folders of OmpX 1 149 measurements without chaperones i.e. "xxDeg_0nM_Skp_Trimer" or "xxDeg_0nM_SurA", the "for RASP analysis" folder contains the pre-processed file used for RASP analysis. - In the folders with OmpX+Chaperone measurements, the "FRET_OmpX_Skp_xnM" or "FRET_OmpX_SurA_xnM" are the files used for performing global chi-square minimization routine to obtain the thermodynamic parameters. All measurements were done in a buffer containing 20 mM TrisHCl and 150 mM NaCl, pH 8.0, unless otherwise stated. *Each column, in the order as present in the "Settings.txt" file, stands for: - Brd_RR(1) ... lower border of the microtime window for red excitation red emission - Brd_RR(2) ... upper border of the microtime window for red excitation red emission - Brd_GGR(1) ... lower border of the microtime window for green excitation green/red emission - Brd_GGR(2) ... upper border of the microtime window for green excitation green/red emission - threIT ... minimal threshold for inter-photon time burst identification - threITN ... maximal threshold for inter-photon time noise region identification - minPhs ... minimal number of consecutive photons satisfying threIT for burst identification - setLeeFilter ...
